5-Amino-1MQ: Blocking the Enzyme That Tells Fat Cells to Store
Inside every fat cell there is an enzyme quietly doing two things at once, and both of them favour storing energy rather than burning it.
Inside every fat cell there is an enzyme quietly doing two things at once, and both of them favour storing energy rather than burning it.
The enzyme is called nicotinamide N-methyltransferase — NNMT. It takes a methyl group from one molecule and attaches it to another, and in doing so it drains two of the cell’s most important metabolic currencies simultaneously. One of them is the raw material for NAD+. The other is the cell’s universal methyl donor.
NNMT is found in unusually high concentrations in adipose tissue, and its expression correlates positively with both obesity and insulin resistance. The more of it there is, the more the metabolic balance tips toward storage.
5-Amino-1MQ is a small molecule designed to stop it. In cultured fat cells at 60 micromolar it reduced lipid accumulation by 70%. In obese mice over eleven days it produced a roughly 35% reduction in fat pad mass.
Those are preclinical figures, and there are no human trials. But the mechanism is unusually well defined, and it works on a pathway that most metabolic research compounds never touch.
5-Amino-1MQ is supplied for laboratory use only — not a medicine, not approved for human consumption, and not sold for any clinical purpose.
First, an Honest Clarification
5-Amino-1MQ is not a peptide. It is worth saying plainly, because it is routinely listed alongside research peptides and frequently described as one.
Its full name is 5-amino-1-methylquinolinium. It is a small-molecule methylquinolinium derivative — a compact aromatic ring system with a positively charged nitrogen, not a chain of amino acids. It has no sequence, no peptide bonds and no molecular weight in the range peptides occupy.
It appears in peptide catalogues because it addresses a metabolic pathway that peptides largely do not reach. That is a sensible reason to stock it and a poor reason to mislabel it.
The distinction matters practically, too. Small molecules are generally more membrane-permeable than peptides, more stable in storage, and not vulnerable to the proteases and oxidation pathways that dominate peptide handling. Nearly every rule about storing peptides exists because peptides are fragile in ways this compound is not.
What NNMT Actually Does
NNMT is a 29 kDa S-adenosylmethionine-dependent enzyme. It catalyses one reaction, and the reaction has two inputs and two outputs.
It takes nicotinamide and SAM. Nicotinamide — vitamin B3 in one of its forms — and S-adenosylmethionine, the cell’s principal methyl donor.
It produces 1-methylnicotinamide and SAH. The methylated nicotinamide, which is then excreted from the body, and S-adenosylhomocysteine, the spent form of the methyl donor.
The enzyme’s three-dimensional structure has been resolved as a ternary complex bound to both SAH and nicotinamide, with aspartate 197 and tyrosine 20 identified as key residues in catalysis. This is a well-characterised enzyme, not a speculative target.
Now the consequence, which is where it becomes interesting.
The NAD+ problem
Nicotinamide is a precursor for NAD+ biosynthesis. Cells recycle it through what is called the salvage pathway — recovering nicotinamide and rebuilding NAD+ from it rather than synthesising the whole molecule from scratch, which is expensive.
NAD+ matters enormously. It is the cofactor that drives oxidative metabolism, and it is required by the sirtuins, a family of enzymes central to metabolic regulation and cellular stress responses.
When NNMT methylates nicotinamide, that nicotinamide is gone. Methylated and destined for excretion, it cannot re-enter the salvage pathway. Every molecule NNMT processes is one fewer molecule available to rebuild NAD+.
Work reviewing NAD+ metabolism specifically in white adipose tissue, published in Adipocyte in 2024 by Kwon and Park, confirmed that NAD+ availability in fat cells directly influences metabolic programming. This is not a peripheral effect.
The methyl donor problem
The second consequence runs in parallel. Every reaction NNMT catalyses consumes one molecule of SAM.
SAM is the cell’s universal methyl donor, used in DNA methylation, histone methylation, and a wide range of other reactions. Depleting it has consequences reaching into epigenetic regulation — which means NNMT activity influences gene expression indirectly, by changing the availability of the currency that methylation reactions require.
So a single enzyme, running continuously in fat tissue, drains both the NAD+ salvage substrate and the methyl donor pool at the same time.
Why Adipose Tissue Specifically
NNMT is present in many tissues — liver most abundantly of all — so a fair question is why adipose tissue became the focus.
Part of the answer is the correlation. NNMT expression in fat tissue tracks with obesity and insulin resistance, which makes it a candidate rather than merely a bystander. An enzyme whose levels rise alongside a condition is worth investigating in either direction: as a cause, or as a consequence worth reversing.
The more interesting part concerns what fat cells do with NAD+.
Adipocytes are not passive storage containers. They are metabolically active, and the balance between storing lipid and oxidising it is regulated rather than fixed. NAD+-dependent sirtuins sit close to the centre of that regulation, influencing which genes are expressed and how mitochondria behave.
A tissue that both expresses NNMT heavily and depends on NAD+-mediated regulation is therefore unusually sensitive to the enzyme’s activity. Draining the NAD+ salvage substrate in a cell type where sirtuin signalling determines storage versus oxidation is a mechanism with a direct route to the observed phenotype.
The 3T3-L1 findings support that reading. The compound suppressed lipid accumulation in cells that were differentiating — being driven from pre-adipocyte to mature adipocyte by adipogenic factors. It was not just reducing storage in established fat cells; it was interfering with the programme that creates them.
Separate work examining adipocyte differentiation-related transcription factors alongside lipid metabolism genes and adipokine expression points the same way. The effect appears to be on the differentiation programme rather than solely on lipid handling downstream of it.
The Compound That Blocks It
5-Amino-1MQ was developed as a probe molecule to test what happens when NNMT is pharmacologically inhibited rather than genetically knocked down.
Its quinolinium scaffold is designed to bind the NNMT active site specifically, leaving other SAM-dependent enzymes largely unaffected at the concentrations studied. That selectivity is not a trivial achievement — SAM-dependent methyltransferases are a large family, and a non-selective inhibitor would produce effects across dozens of unrelated pathways.
The second critical property is membrane permeability. NNMT operates intracellularly, so an inhibitor that cannot cross the cell membrane is useless regardless of how well it binds. In vitro studies established that 5-Amino-1MQ has high cell permeability alongside its enzyme selectivity — the combination that made in vivo work possible.
Inhibiting NNMT should, in principle, do the reverse of what the enzyme does: preserve nicotinamide for NAD+ salvage, and preserve SAM for methylation reactions. Both pools rise rather than fall.

What the Preclinical Work Found
The evidence has a clear structure, running from cell culture through to animal models.
The genetic groundwork
The foundation is a 2014 Nature paper by Kraus and colleagues establishing that NNMT knockdown protects against diet-induced obesity. Removing the enzyme genetically produced the metabolic phenotype the pharmacology later aimed at.
Subsequent work in Nature Chemical Biology in 2015 reported that NNMT regulates hepatic nutrient metabolism through Sirt1 protein stabilisation — connecting the enzyme directly to sirtuin function, which is what the NAD+ argument would predict.
Cell culture: lipid accumulation
The 3T3-L1 cell line is the standard in vitro model for studying fat cell formation — pre-adipocytes that can be driven to differentiate into mature adipocytes with the right signals.
Treating differentiating 3T3-L1 cells with 5-Amino-1MQ in adipogenic media produced concentration-dependent inhibition of lipid accumulation, with strong statistical significance.
The specific figures from the patent literature:
30 micromolar reduced lipogenesis by 50%.
60 micromolar reduced lipogenesis by 70%.
Importantly, cell viability was only slightly reduced even at the highest concentration tested — meaning the effect on lipid accumulation was not simply a consequence of killing the cells, which is the obvious alternative explanation and the one a careful reader should look for.
Animal models: the eleven-day study
Because the in vitro data showed high cell permeability, enzyme selectivity and cell culture efficacy, a sub-chronic proof-of-concept study was conducted in diet-induced obese mice maintained on a high-fat diet.
The protocol: 20 mg/kg of 5-Amino-1MQ, subcutaneously, three times daily, for 11 days.
The results:
Progressive loss of body weight across the treatment period compared with controls.
A roughly 35% decrease in epididymal white adipose tissue mass, at P less than 0.001.
More than 30% decrease in adipocyte size on histological analysis, at P less than 0.05.
More than 40% decrease in adipocyte volume.
That last pair of findings is worth dwelling on. The fat pads did not merely weigh less — the individual fat cells within them were measurably smaller. That is consistent with reduced lipid storage per cell rather than with cell death, and it matches what the 3T3-L1 work predicted.
Diet combination
A 2022 study in Scientific Reports, indexed under PMC8748953, examined NNMT inhibition alongside dietary change in diet-induced obese mice.
The headline finding: 5-Amino-1MQ combined with a low-fat diet promoted dramatic whole-body adiposity and weight loss, rapidly normalising these measures to age-matched lean animals — while switching to the low-fat diet alone was unable to restore those measures in the same timeframe.
The study’s actual purpose was to examine the cecal microbiome, testing whether treated animals resembled lean counterparts microbiologically as well as metabolically. But the weight finding is the more striking one for metabolic research: the compound did something diet alone did not, within the same window.
Autophagy and transcription factors
Separate work indexed under PMC9276046 examined NNMT’s effects on lipid accumulation, triglyceride content, adipocyte differentiation-related transcription factors, lipid metabolism genes, adipokine expression and autophagy in 3T3-L1 adipocytes — broadening the mechanistic picture beyond the NAD+ and SAM axes alone.
Designing Research With 5-Amino-1MQ
Several practical points follow from the mechanism, and most differ from how peptide protocols are usually built.
Work in the micromolar range. The effective concentrations in cell culture were 30 and 60 μM — three to six orders of magnitude above where many research peptides operate. Protocols adapted from peptide work will be far too dilute.
Measure the pathway, not just the outcome. The informative endpoints sit upstream of weight or lipid content: intracellular NAD+ levels, SAM and SAH concentrations, and 1-methylnicotinamide output. If NNMT is being inhibited, 1-methylnicotinamide should fall while nicotinamide and NAD+ rise. That is a direct test of mechanism rather than an inference from phenotype.
Include a viability control. The patent data specifically reported that 3T3-L1 viability was only slightly reduced at the highest concentration. That control matters, because reduced lipid accumulation in dying cells is not the same result as reduced lipid accumulation in healthy ones. Any replication should measure both.
Use differentiating cells, not just mature ones. The strongest cell culture effects came from pre-adipocytes undergoing differentiation in adipogenic media. Testing on already-differentiated adipocytes asks a different question and may give a weaker answer.
Check selectivity at your concentration. Selectivity against other SAM-dependent methyltransferases was established at the concentrations studied. Work well above that range should not assume the qualifier still holds.
Account for dosing frequency in vivo. The animal protocol used three daily subcutaneous administrations, which implies fairly rapid clearance. A once-daily design is not a reduced version of the same experiment.
Consider the diet interaction. The Scientific Reports work found that the compound combined with a low-fat diet achieved what diet alone did not in the same timeframe. Diet is therefore a variable rather than a background condition, and holding it constant across arms matters more than usual.
Beyond Fat: The Oncology Thread
NNMT research extends well outside metabolism, and it is worth knowing because it says something about the enzyme’s significance.
NNMT is overexpressed in a variety of human cancers and has been investigated as an anti-cancer target in its own right.
Work indexed under PMC5216764 examined NNMT in colorectal cancer cells, reporting that the enzyme enhanced resistance to 5-fluorouracil chemotherapy through inhibition of the ASK1-p38 MAPK pathway. Cells with high NNMT expression were less sensitive to the drug; knocking NNMT down reversed that.
More recent work indexed under PMC12650368 tested 5-Amino-1MQ directly — alongside two other NNMT inhibitors — on osteosarcoma and Merkel cell carcinoma cell lines, examining chemosensitivity.
For metabolic researchers this matters in two ways. It means the enzyme is broadly consequential rather than narrowly adipose-specific. And it means a compound inhibiting it may have effects beyond the intended tissue, which is a consideration for study design rather than a problem.
How This Differs From Taking NAD+ Directly
The obvious question: if the mechanism runs through NAD+ availability, why not simply supply NAD+ or one of its precursors?
The answer identifies a genuine mechanistic distinction.
Direct NAD+ supplementation provides the cofactor. More substrate enters the system.
NNMT inhibition works upstream, preventing nicotinamide depletion. Rather than adding more, it stops the existing pool being drained.
Those are different interventions with different consequences. And there is a second layer: NNMT inhibition also affects SAM availability and downstream epigenetic marks — a mechanistic dimension that direct NAD+ supplementation does not engage at all.
So the two approaches are complementary rather than interchangeable, which makes comparative work across both genuinely informative. NAD+ and 5-Amino-1MQ address the same axis from opposite ends.
Where the Evidence Stops
Being clear about this is more useful than enthusiasm.
There are no published human trials. The evidence base for 5-Amino-1MQ consists entirely of animal studies and in vitro work. No human data exists to confirm that the preclinical findings translate.
Much of the compound-specific work is cell culture. Most published work on 5-Amino-1MQ itself, as distinct from NNMT knockdown studies, has been conducted in cell culture systems. The genetic knockdown literature is broader than the pharmacological literature.
The in vivo study was short and intensive. Eleven days at 20 mg/kg three times daily is a demanding protocol. It establishes proof of concept; it does not establish what happens over longer periods or at lower frequencies.
Selectivity is concentration-dependent. The compound leaves other SAM-dependent enzymes largely unaffected at the concentrations studied. That qualifier is in the source literature and should stay attached.
What remains is a well-defined enzyme target with a resolved crystal structure, a selective and membrane-permeable inhibitor, consistent dose-dependent effects across cell and animal models, and a mechanism that makes sense in both directions. For a research compound, that is a solid position — and the absence of human data is a gap rather than a contradiction.
Related Compounds Worth Studying Alongside
5-Amino-1MQ occupies an unusual position, which makes its comparators unusually varied.
The NAD+ axis. NAD+ is the most direct comparison — same pathway, opposite intervention point, as described above. Running both distinguishes substrate availability from salvage preservation.
Mitochondrial compounds. Since NAD+ drives oxidative metabolism, compounds acting on mitochondrial function are natural companions. MOTS-c is a mitochondrially-encoded peptide studied in metabolic regulation, and SS-31 targets mitochondrial membranes specifically. Methylene Blue is another non-peptide small molecule studied for mitochondrial electron transport — a useful structural parallel, since it too sits in a peptide catalogue without being a peptide.
Adipose and metabolic compounds. AOD-9604 is a fragment of growth hormone isolating its lipolytic function, approaching fat metabolism from an entirely different direction. L-Carnitine is involved in fatty acid transport into mitochondria. Tesamorelin and the wider metabolic research range cover adjacent territory, and the metabolism and energy research bundle groups several for comparative protocols.
Growth hormone secretagogues. Because growth hormone influences lipolysis, Ipamorelin, GHRP-2 and CJC-1295 with Ipamorelin appear in overlapping research.
Longevity compounds. Since sirtuins sit downstream of NAD+ availability, Epitalon and Pinealon are relevant — the latter particularly so, since its mapped gene targets include SOD2 and the PPAR receptors, connecting to the same metabolic and antioxidant territory.
Antioxidant compounds. Glutathione is studied in oxidative stress contexts that intersect with mitochondrial metabolism.
Handling and Storage
Because 5-Amino-1MQ is a small molecule rather than a peptide, its handling requirements are meaningfully less demanding.
There is no peptide backbone to hydrolyse, no oxidation-prone amino acid residues, no disulfide bond, and no risk of the aggregation that affects peptides in solution. The failure modes that dominate peptide storage largely do not apply.
That said, good practice still helps:
Store as supplied at −20°C, protected from light, for long-term stability.
Bring to room temperature before opening to avoid condensation onto the material.
Keep solutions refrigerated and use within a defined window. As an aromatic compound with a charged nitrogen, it is chemically robust, but solution stability is always shorter than dry stability.
Note the concentrations involved. The effective range in cell culture work is micromolar — 30 to 60 μM — which is considerably higher than the nanomolar and picomolar ranges typical of peptide work. Stock preparation should account for that, and the calculator handles the arithmetic. Full guidance is in the storage information guide.
Verification and Certificates
Small-molecule verification differs from peptide verification in emphasis.
There is no sequence to confirm, no deletion sequences to separate, and no terminal modifications to check. What matters is chemical identity and purity — that the compound is 5-amino-1-methylquinolinium and not a related quinolinium derivative or an incompletely aminated precursor.
HPLC establishes purity by separating the target from related substances. Mass spectrometry confirms molecular weight, and for a compact aromatic molecule the mass is distinctive enough that identity confirmation is straightforward.

What is 5-Amino-1MQ?
5-amino-1-methylquinolinium, a membrane-permeable selective small-molecule inhibitor of nicotinamide N-methyltransferase. It is a methylquinolinium derivative, not a peptide.
What is NNMT?
Nicotinamide N-methyltransferase, a 29 kDa SAM-dependent enzyme that methylates nicotinamide to produce 1-methylnicotinamide and SAH. It is found in high concentrations in adipose tissue, and its expression correlates positively with obesity and insulin resistance.
How does blocking NNMT affect fat storage?
NNMT consumes nicotinamide, which is a precursor for NAD+, and SAM, the cell’s methyl donor. Inhibiting it preserves both pools. Higher NAD+ availability activates NAD+-dependent enzymes including sirtuins, shifting adipocyte metabolism toward energy expenditure rather than storage.
Is 5-Amino-1MQ a peptide?
No. It is a small-molecule quinolinium derivative with no amino acid sequence and no peptide bonds. It appears in peptide catalogues because it addresses a metabolic pathway few peptides reach.
What did the animal study show?
In diet-induced obese mice given 20 mg/kg subcutaneously three times daily for 11 days, treatment produced progressive body weight loss, a roughly 35% decrease in epididymal white adipose tissue mass, more than 30% decrease in adipocyte size, and more than 40% decrease in adipocyte volume.
What concentrations were effective in cell culture?
In differentiating 3T3-L1 pre-adipocytes, 30 μM reduced lipogenesis by 50% and 60 μM reduced it by 70%, with only slight reduction in cell viability at the highest concentration tested.
Are there human trials?
No. The published evidence base consists entirely of animal studies and in vitro work. No human data exists to confirm whether the preclinical findings translate.
How does it compare with taking NAD+?
NAD+ supplementation supplies the cofactor directly. NNMT inhibition works upstream by preventing nicotinamide depletion, preserving the salvage pathway. It also affects SAM availability and downstream epigenetic marks, which NAD+ supplementation does not.
Why does membrane permeability matter?
NNMT operates inside the cell. An inhibitor unable to cross the cell membrane cannot reach it regardless of binding affinity. High cell permeability alongside enzyme selectivity is what made in vivo studies possible.
Is NNMT studied outside metabolism?
Yes. It is overexpressed in a variety of human cancers and investigated as an anti-cancer target, including work on 5-fluorouracil resistance in colorectal cancer and chemosensitivity in osteosarcoma and Merkel cell carcinoma.
How should it be stored?
As supplied at −20°C, protected from light. As a small molecule it lacks the peptide backbone, oxidation-prone residues and aggregation risks that make peptide storage demanding, so it is considerably more robust.
One Enzyme, Two Currencies
What makes NNMT an interesting target is that it drains two things at once.
Every reaction it catalyses removes a nicotinamide molecule from the NAD+ salvage pathway and spends a molecule of SAM. One enzyme, running continuously in the tissue where it is most concentrated, steadily reducing both the cofactor that drives oxidative metabolism and the donor that drives methylation.
Block it and both pools recover. That is the whole hypothesis, and the preclinical data follows it closely — 50% and 70% reductions in lipogenesis at 30 and 60 micromolar in cell culture, then 35% less fat pad mass and measurably smaller individual adipocytes in obese mice over eleven days.
What is missing is human evidence, and that absence is real rather than a technicality. But the enzyme’s structure has been resolved down to the catalytic residues, the inhibitor is selective and membrane-permeable, and the mechanism explains the results rather than being inferred from them.
For research on adipose metabolism, it is the most characterised small-molecule NNMT tool compound available — and it reaches a pathway that the peptides dominating metabolic research do not touch.
