Background

About Selank Peptide UK

Selank is a synthetic heptapeptide derived from a sequence related to the naturally occurring immunomodulatory peptide tuftsin. It is used in neurobiological research examining neurotransmitter systems, stress responses, cognition and interactions between neural and immune signalling.

Also known as: Selank peptide, Selank acetate, TKPRPGP.

Research

Selank Research

Selank research has explored GABAergic signalling, gene-expression changes, serotonergic and other neurotransmitter pathways, learning and memory models, stress responses and neuroimmune regulation (Learn more). Its literature is notable for spanning both neurochemical and immune-related mechanisms, making the peptide relevant to research on the interface between stress, cognition and signalling, alongside related nootropic and neuro-research peptides such as Semax and DSIP.

A modulator at the GABA receptor, not a direct key

Clinical reports that Selank resembles benzodiazepines prompted Russian biochemists to test the idea directly in rat brain membranes. Selank altered the binding of tritiated GABA in the manner of a positive allosteric modulator, changing how GABA itself attaches rather than occupying the GABA site (Learn more). When it was combined with benzodiazepines the binding changes were not simply additive, and Selank could block the modulating effect of diazepam and of the antipsychotic olanzapine. The authors concluded that the sites involved are probably different but may partly overlap.

Route of delivery seems to matter as well. In BALB/c mice, Selank given by intraperitoneal injection raised the number of GABA-receptor binding sites labelled by [3H]SR 95531 in the frontal cortex by 38%, with no change at hippocampal NMDA receptors. Intranasal delivery did the reverse, increasing binding of the NMDA-channel marker [3H]MK-801 by 23% while leaving GABA sites untouched.

Two mouse strains, two very different responses

Much of Selank's early pharmacology was built on a comparison between two inbred mouse strains. BALB/c mice explore little and react passively to stress, while C57BL/6 mice are far more active in the same tests. In open-field and elevated plus maze experiments, Selank's anxiolytic-like behavioural effects showed up in BALB/c animals but were weak or absent in C57BL/6.

The opioid system adds a twist. Pretreatment with naloxone, which blocks opioid receptors, weakened the response to Selank in BALB/c mice yet strengthened it in C57BL/6 mice (Learn more). The research team read this reversal as evidence of an additional opioid-linked target for the peptide, and as a sign that the starting emotional state of the animal shapes what Selank does.

Serotonin and monoamine turnover in rodent brain

Brain chemistry measurements help explain those strain differences. Even before treatment, BALB/c mice carried more serotonin and more of its breakdown product 5-HIAA in the hippocampus than C57BL/6 mice. Selank then lowered hippocampal serotonin and 5-HIAA in BALB/c animals while leaving C57BL/6 levels alone, raised noradrenaline in the hypothalamus of both strains, and pushed the dopamine metabolites DOPAC and HVA in opposite directions: up in the frontal cortex and hippocampus of C57BL/6 mice, down in BALB/c (Learn more).

Rat studies add a time dimension. In Wistar rats trained in a food-reward task, a single Selank injection given partway through training activated serotonin metabolism in the hypothalamus and caudal brainstem for between 30 minutes and two hours. Retention was tested at 24 hours, 7 days and 30 days, and the treated rats kept a more stable memory trace across that period.

Tuftsin heritage: spleen genes, cytokines and interferon

Tuftsin is an immune peptide, so it is no surprise that Selank has been examined in immune tissue. In mouse spleen, a single injection changed the expression of 34 out of 84 inflammation-related genes, covering chemokines, cytokines and their receptors, at 6 and 24 hours (Learn more). The transcription regulator Bcl6, important in immune-cell development, responded to Selank and to each of the fragments tested. A follow-up time course found complement C3 mRNA down about threefold only 30 minutes after injection, with Il2rg, Casp1 and Xcr1 shifting within the first 90 minutes.

Influenza experiments produced one of the most striking results. Against an H3N2 influenza A strain, Selank added to cell cultures 24 hours before infection completely suppressed viral reproduction, and in infected mice survival was highest when the peptide was given ahead of infection. In those mice it induced interferon-alpha gene expression without changing IL-4, IL-10 or TNF-alpha.

Hippocampal transcriptome and BDNF work in rats

Microarray profiling of the rat hippocampus found 36 genes changed more than twofold after a single intranasal application of Selank, and 20 after a course. Most of them encode proteins of the plasma membrane, many spanning it, which led the authors to propose that Selank influences ion balance in hippocampal cells and, through it, ion-dependent processes such as memory formation in rodents (Learn more).

BDNF has entered the Selank literature through the hippocampus too. Intranasal Selank was reported to regulate hippocampal BDNF expression in rats, and in a long-term model in which rats drank 10% ethanol as their only fluid for 30 weeks, Selank prevented the ethanol-linked rise in BDNF content in both the hippocampus and the frontal cortex. In the same model it countered deficits in an object recognition test during withdrawal.

Questions

Selank Frequently Asked Questions

About Selank6 questions
What is Selank?

Selank is the synthetic heptapeptide Thr-Lys-Pro-Arg-Pro-Gly-Pro (TKPRPGP), with the formula C33H57N11O9, a molecular weight of 751.89 and CAS number 129954-34-3. Its first four residues reproduce the immune peptide tuftsin and the last three add a glyproline tail. It is also listed as Selank peptide or Selank acetate, and Crown Peptides supplies 5mg and 10mg vials.

Does Selank bind the GABA site itself?

Radioligand work suggests not. In rat brain membranes, Selank changed how tritiated GABA bound, acting as a positive allosteric modulator: a substance that tunes the receptor's response to GABA from a separate site. That places it closer in mechanism to benzodiazepines than to GABA, although its site does not appear identical to theirs.

What happens when Selank meets diazepam in laboratory studies?

The two interact in ways that are not simply additive. In membrane binding assays, Selank blocked diazepam's modulating effect on GABA binding. In unstressed rats, a course of either compound nudged elevated plus maze measures in an unfavourable direction, but the shift was smaller with Selank, one of several hints that the two act through related yet distinct mechanisms.

Why do so many Selank studies compare BALB/c and C57BL/6 mice?

The two strains differ in baseline anxiety-like behaviour and in brain monoamine levels, which makes them a natural test of selectivity. Selank's behavioural and neurochemical effects repeatedly appeared in the passive, high-anxiety BALB/c strain and were weak or absent in C57BL/6 mice, a pattern the researchers interpret as an effect that depends on the animal's initial state.

What did Selank do to brain serotonin in rodent studies?

The answer depended on the animal. In BALB/c mice, hippocampal serotonin and 5-HIAA fell after Selank, while C57BL/6 mice showed no change. In Wistar rats, a single injection during training activated serotonin metabolism in the hypothalamus and caudal brainstem for up to two hours, a finding the authors tied to the peptide's effect on memory retention in that task.

Why was Selank tested against a protein-synthesis blocker?

Actinomycin D halts RNA synthesis and, with it, the new protein production that long-term memory consolidation requires in animals. Researchers used it to disrupt learning in rats trained to find the safe exit in a 16-door maze, and Selank prevented or offset that disruption while shortening the time the rats needed to learn the task.

Immune and gene-expression research5 questions
Why is the mouse spleen such a common tissue in Selank research?

The spleen is packed with lymphocytes and macrophages, so it gives a direct readout of immune gene activity after a single treatment. Because Selank descends from tuftsin, which was first characterised through its action on immune cells, the Moscow group used spleen panels of chemokine, cytokine and receptor genes to check whether that immune heritage survived in the longer peptide.

What is the Gly-Pro fragment, and why do Selank papers keep testing it?

Gly-Pro is the dipeptide at the very end of Selank. Antiviral screening identified it as the smallest fragment that kept antiviral activity, so the group calls it the Selank pharmacophore. In mouse spleen, Gly-Pro reproduced many of the gene-expression changes seen with the full peptide, including a similar fall in C3 mRNA, which makes it a handy comparison for separating the roles of the two halves.

What is known about Selank and interleukin-6?

The data are mixed but interesting. In cultured blood cells from patients with depression, Selank at 10-7 M completely suppressed IL-6 gene expression, an effect not seen in cells from healthy donors, yet IL-6 protein measured in the patients' cultures rose.

Has Selank been tested for effects on blood clotting?

It has, using thromboelastography, which records how a clot forms and strengthens over time. Selank, the related glyproline His-Phe-Arg-Trp-Pro-Gly-Pro and the tripeptide Pro-Gly-Pro all shifted clotting parameters towards slower, weaker coagulation compared with controls, and Selank showed the strongest anticoagulant effect of the three.

How have researchers tracked Selank after it is administered?

By labelling every residue of the peptide with tritium through high-temperature catalytic isotope exchange, then extracting tissue and separating intact Selank from its breakdown products by chromatography. Because the label sits on all seven residues, nearly every possible fragment can be measured at once, and the method was used both for degradation pathways in blood plasma and for brain concentrations after intranasal delivery in animals.

Lab notes3 questions
How does Selank's run of basic residues show up in analysis?

At neutral pH Selank carries three positive charges, from its amino terminus, lysine and arginine, and it has no acidic side chains to offset them. In electrospray mass spectrometry it therefore appears readily as doubly and triply charged ions, near m/z 376.7 and 251.5, as well as the singly charged ion at about 752.4 (monoisotopic mass 751.43). The same positive charge can make it cling to glass and some plastics in very dilute solutions.

Should a Selank project start with one vial or a full box?

A single 5mg vial is a sensible way to set up and validate an assay. Once a protocol is fixed, the 10mg strength and the five- or ten-vial boxes reduce the cost per milligram and keep one batch running through a whole series of binding, behaviour or gene-expression experiments, which helps when results from different weeks need to be compared.

Is Selank also available pre-mixed with Semax?

It is. Crown Peptides also sells a Semax + Selank blend containing 5mg of each peptide per vial, for projects that want both in a single preparation. The single Selank vial remains the better choice when its own effects need to be isolated.
